How do you know you maintain the TME?

We confirm the preservation of the tumor microenvironment (TME) by functionally measuring cytokines and chemokines that indicate intact immune�tumor interactions. After ex vivo treatment with IO therapies, Elephas assesses the dynamic secretion of key immune mediators, such as IFN-?, CXCL9, CXCL10, and others, which are produced only when viable immune and stromal networks remain functional. The balance between inflammatory and immunosuppressive signals provides strong evidence that the TME architecture and signaling are maintained. At the end of each assay, a CD3/CD28 stimulation control is applied to the same fragments (the elive Method) to confirm that T cells remain viable and capable of activation. Samples that fail to respond to this control are flagged as non-informative and excluded from analysis. Together with viability assays, histopathologic review, and multiplex cytokine profiling, these steps ensure that the elive maintains and verifies the integrity and functional responsiveness of the live TME.
